Tsukune Aono is an average teenager who is unable to enroll in a local high school due to his poor grades. As a final attempt to educate him, his parents enroll him in a private school called Youkai Academy, which he discovers is a boarding school for monsters. The school teaches monsters how to coexist with people, including disguising them as themselves, but all real people found on campus must be killed. Although he fears for his life, Tsukune befriends a beautiful vampire girl named Moka Akashiya, who enjoys his company and especially the taste of his blood. During a fight with a school bully, Tsukune accidentally removes the rosario from Moki’s neck and discovers that she is transforming into a powerful vampire with a completely different personality.
Because of Moka, Tsukune decides to stay with the youkai. He is friends with former student antagonists, including the succubus Kuruma Kurono and the prodigy Yukari Sendo. They join the school’s newspaper club and end up fighting dozens of students and teachers who use their monstrous powers to intimidate or control one or more members for their own selfish or destructive purposes. During a club trip to the world of people, they meet the witch Ruby Tojo, who protects the sunflower field from destruction by the developers.
They meet Mizore Shirayuki, the snow fairy’s apprentice, who becomes Tsukune’s “pursuer”. The club is disturbed by a group of “monsters”. Ruby, recovering from her injuries, joins the school team as Deputy Principal. An evil group called “Anti-School Children” threatens to destroy the barrier between the human world and the monster worlds. When Tsukune’s cousin manages to infiltrate the school grounds, the club struggles to keep the monster school a secret. When the school is temporarily closed due to damage, Tsukune returns home to the human world only for Moka, Kurumu, Yukari, and Mizore to follow him and hide in his room.
Rosario + Vampire: Season 2 registers Moka’s younger half-sister Koko Shuzen. She, Ruby and Mizore join the Newspaper Club. During an excursion to the homeland of Mizore, they learn about a dangerous organization called “Fairy Tale”, which seeks to destroy the human world. The newspaper club visits the human world, where they meet up with San Otonashi, a former member of the club, and run into trouble with opponents from the 7th branch of the Fairy Tale. Chinese translation student Fanfan Huang tries to recruit Tsukune to join his mafia family. When Moka spends her school day as her inner self, she cannot go back to the seal, so the newspaper club heads to China to fix her Rosario.
In Hong Kong, Tsukune and the gang discover the truth about Moki’s identity: she was drenched in the blood of the First Ancestor by her mother Akasha Bloodriver, who sacrificed herself to protect them from Alucard, the first First Ancestor of the vampire. Moka’s older half-sister, Aqua, infiltrates Huang’s estate and captures Moka to take him to Fairy Tale, which suspects Moka is the key to Alucard’s resurrection. While his friends train their powers, Tohofuhai alters his Tsukune body to cope with the upcoming trials.
Tsukune and the gang infiltrate the Hanging Garden: a heavenly fortress that serves as the Fairy Tale’s headquarters. They face opposition from their leader and matriarch of the Shuzen family, Gyokuro Shuzen, who sends the leaders of the Fairy Branch after them on a series of battles. The group eventually reaches Moki, but encounters her half-sisters Aqua and Kalua. Gyokuro grabs Moki’s rosario to control Alucard. A character named The Masked King turns out to be a clone of the original vampire form Alucard. He reveals that he has manipulated both Fairy Tale and her opposition to determine if Rosario can control him, and then merges with his monster, which is rampaging in Tokyo.
Moka is mortally wounded, but Tsukune revives her, removing all of her sacred castles and transforming into the vampire of the First Ancestor. They beat Alucard, but he comes to life and reveals that he is hatching eggs in the major cities of Japan. Tsukune’s friends and allies, along with two other Dark Lords, team up to fight Alucard and his clones. Tsukune puts Moki’s rosario on Alucard, which awakens Akasha in his monster body. Akasha tells Alucard to stop fighting and they cast a self-destruction spell. Now that the human world is aware of the existence of monsters, coexistence has become even more difficult; however, Tsukune is confident that he and his friends can do it.
